Danocrine (Danazol) Information

Danocrine, with the active ingredient Danazol, is a synthetic derivative of the male hormone testosterone. It is available in capsule form, typically containing 100mg or 200mg of Danazol.

Indications

Danocrine is primarily prescribed for the treatment of:

  • Endometriosis: To reduce or eliminate menstrual pain, abnormal uterine bleeding, and pelvic masses associated with endometriosis.
  • Fibrocystic Breast Disease: To relieve pain and tenderness in the breasts.
  • Hereditary Angioedema: To prevent attacks of swelling in patients with this condition.
  • Idiopathic Thrombocytopenic Purpura (ITP): In certain cases where other treatments have failed.

Mechanism of Action

Danazol exerts its effects by acting on the hypothalamic-pituitary-gonadal axis. It suppresses the release of gonadotropins (luteinizing hormone [LH] and follicle-stimulating hormone [FSH]) from the pituitary gland. This reduction in LH and FSH leads to decreased production of ovarian steroid hormones (estrogen and progesterone). In endometriosis, this hormonal suppression leads to a regression of endometrial tissue. In fibrocystic breast disease, it reduces estrogen levels, alleviating breast symptoms. For hereditary angioedema, it is thought to increase the levels of C1 inhibitor protein, thereby preventing swelling attacks.

Safety Profile

Benefits: Effective in managing symptoms of various gynecological and hormonal conditions.

Side Effects: Common side effects may include weight gain, fluid retention, acne, changes in libido, hot flashes, and mood changes. Less common but serious side effects can include liver function abnormalities, thromboembolic events, and changes in blood cell counts. Women of childbearing potential should use non-hormonal contraception and undergo pregnancy tests before and during treatment due to the risk of virilization in a female fetus.

Contraindications: Danocrine is contraindicated in individuals with undiagnosed abnormal genital bleeding, pregnant or breastfeeding women, and those with hypersensitivity to danazol or any of its components. Caution is advised in patients with liver, kidney, or heart disease.

Warnings: Regular monitoring of liver function, blood cell counts, and lipid profiles is recommended during long-term therapy. Patients should be closely monitored for signs of thromboembolism.

Frequently Asked Questions

Q1: How long does it take for Danocrine to work?
A1: The onset of therapeutic effect varies. For endometriosis, significant pain relief may take several weeks to months. For ITP, improvements in platelet counts can sometimes be observed within weeks.

Q2: Can I drink alcohol while taking Danocrine?
A2: Alcohol consumption should be discussed with your doctor. Danazol can affect liver function, and alcohol may exacerbate this risk. It's generally advisable to limit or avoid alcohol.

Q3: What happens if I miss a dose of Danocrine?
A3: If you miss a dose, take it as soon as you remember unless it is almost time for your next dose. Do not take a double dose to make up for a missed one. Consult your healthcare provider if you are unsure.

Q4: Is Danocrine safe for long-term use?
A4: Danocrine can be used for extended periods under medical supervision. However, regular monitoring of liver function, lipid levels, and other parameters is essential to manage potential long-term side effects.
